Besides the obviously challenging Les Miz, A Chorus Line, etc...
The Mystery of Edwin Drood (a sophisticated score and a difficult show to pull off on the whole, plus lots of roles!)
Big: the musical (someone else suggested this... i like this idea)
The Who's TOMMY (careful no one blows out their voice)
Once On This Island (a school with a fabulous theatre program did this nearby, they had almost no African-Americans in the school, but they made a few adjustments and the show turned out beautifully, one of the best high school productions i've ever seen)

If you want hard I'd definately suggest "The Pirates of Penzance". The set construction needs to be top notch as a pirate ship is needed and then there's the house, or tower, or whatever you want to call it for the second act. The vocals are really, really demanding because it is an operetta and there's a huge cast. I was involved in the show last year and trust me, it's difficult.
Also, anything Sondheim is pretty difficult to do. "Into the Woods" has numerous sets and Sondheim's vocals are demanding.
