I'm getting in line on Thursday to get tickets to "Two Gentlemen of Verona". Which is the best line to get in and what time should I be there to assure myself seats? Thanks!

by 9 am....people have been getting there at 4:30 in the morning. the tickets are going very quickly, as its the last week.

The line at the Public is generally shorter, though you still may have to be there by 9-ish to ensure tix.
You might also try the cancellation line in the Park if that's more convenient. It depends, but generally if you're in line 6pm, you'll get in (it's only one ticket per person though, so if you want to go with a friend, he/she has to be there in line with you). Somedays the last person to get in arrived at 5:30, other times you can get there as late it 6:45, so it's hard to predict.

I've seen the show twice. Once I arrived at the Delacorte at 8 am (I was fourth on line) and once I got on the cancellation line at 6:00 pm. The cancellation line gave me a better seat.
If you definitely want seats, go to the Delacorte. I would be on line before 10 am to guarantee a seat.

On a Thursday, 10AM would be a safe bet.
I was in the park this morning and at 10:00 AM, there wasn't a very long line. But on a weekend, it can be packed. We got there at about 8:30 last weekend and were able to get tickets.
The day I went, I arrived super early at 7:30am and I was glad I did. Half the house was already given to benefactors for an event, so a ton of people who normally would have received tickets were turned away. I was told that as a general rule, arrive before 10am, but the earlier you arrive, the better the seats. I wish I could see it again. I loved every moment. An utter delight from start to finish. The standout performances of Goldsberry, Lewis and Lawrence should not be missed!
A snore??? Not to me. The night I saw it, the audience was cheering and having a great time. "Land of Betrayal", "Bring All the Boys Back Home", "Night Letter", "Love's Revenge", "Thurio's Samba" and "Love Me" all received huge ovations. At the very least, it deserves a new cast recording.
Getting there earlier does not guarantee you better seats. I know from experience. Seating is completely luck of the draw.

My problem with the show, Mister Matt, was mainly the first act. It's very long and FULL of exposition to the point that it becomes a little too much. The second act, however, if you're not bored enough to stick around, is FABULOUS.

Where is the Delacorte and also the Public?

The Delacorte is a short walk from the 81st Street entrance to the Park (take the B or C to 81st).
The Public is on Lafayette next to Astor Place and Cooper Union (it's a huge building and hard to miss). You can take the 6 to Astor Place or the N/R to 8th Street and you're steps away from it.
