I second Dollypop's choice for Mme Armfeldt...Zoe Caldwell !
And please can we not have the cast playing the instruments. Desiree on tuba is just too ridiculous for words, maybe on zither.
Desiree- Glenn Close (whatever, I think she'd be great)
Fredrik- John Dossett
Charlotte- Jan Maxwell
Madame Armfeldt- Angela Lansbury (would she do it? possible but doubtful, but whatever)
Count Magnus- Marc Kudisch
Petra- Donna Lynne Champlin
Anne- Elena Shaddow
Henrik- Shonn Wiley

I'd like to see Donna Murphy as Desiree and Cerveris as Count Carl Magnus

As much as I admire Donna Murphy, I believe she's too young for Desiree, and also this is a role written for a leading lady who was not a singer; when I see Murphy back on a Broadway musical I want it to be in a show where she can show off her brilliant singing.
I believe Meryl Streep is an excellent choice for the role. I also think either Bernadette Peters or Victoria Clark would do justice to Desiree.
Other choices:
Fredrick-James Naughton, Jonathan Pryce.
Anne-Laura Benanti, Lauren Molina, Kelli O'Hara.
Henrik-Matthew Morrison, Steven Sutcliffe.
Count Magnus-Brent Barrett.
Charlotte-Debbie Gravitte, Veanne Cox.
Mme.Armfeldt-Elaine Stritch, Angela Lansbury.
Petra-Tammy Blanchard, Kim Crosby.

Matthew Morrison couldn't sing HEnrik. He just barely made it through Fabrizio. He certainly doesn't have a high B. Apparantly Sondheim believes Danny Gurwin is the best Henrik he's ever seen. I'm not a fan of his upper range but when the creator makes that kind of comment, you listen.
Donna Murphy just turned 47. She's not too young to play Desiree...
Kim Crosby is, however, way too old to play Petra. And Tammy Blanchard could never sing it. Donna Lynne Champlin, though, might bring a kind of course earthiness to the role that would be interesting.
I'd love to see either Victoria Clark or Michele Pawk play Desiree.
uh donna lynn's a little long in the tooth for Petra
I vote for Marin Mazzie for Charlotte.
...but I'm biased.
Although she's a bit too "glam" for it... I think she could tone that down enough to be very convincing, touching (and hilariously bitchy) as the "forgotten wife."
And her "Everyday a Little Death" would rule.
Peter Friedman as Henrik??? maybe 30 years ago...
CLEARLY 30 years ago....I think charlotte's not enough for our dear Miss Mazzie. I wish Emily Skinner looked a little bit more like a Petra lately...

I personally would love to see Michael Rupert as Fredrick...And Danny Gurwin was amazing as Henrik at Kennedy Center, would love to see him reprise it!
I didn't see Danny Gurwin in ALNM, but I would certainly LOVE to. I really do enjoy his high register.
If Meryl Streep signed on as Desiree, I'll bet Miss Mazzie would be happy to join that cast in any part! Even Mala.
But if it's not someone that most actors would kill to work with (like Streep)... you're probably right that Charlotte isn't a big enough part for Marin.
Too bad Marin isn't old enough yet for Desiree! Another 5 to 10 years oughta do it. I'm already looking forward to seeing her play either of the female leads in "Follies"... but that's a while off yet too.

The best Desiree....and Megan Mulally for Petra?
"Miller's Son" needs to be sung by a younger woman who's coming to terms with her future prospects in love and in life.
Megan's been down that road already. It wouldn't work coming from someone her age. Just my two cents.
Petra is a very small part with that wonderful 11 o'clock number. It should go to an up-and-coming unknown.
Unless Streep plays Desiree, and then you might end up with a limited run and an "all star" cast!

They should fly Diana Rigg across the pond to play Desiree.
Marin Mazzie should play Mala! But she needs to play it exactly the same way as Despo.
Even if Emily was "looking more like a Petra these days", you DO know she's older than Donna Lynne, right? Only by a year, but older nonetheless.
I think they're both a little past their prime to play the role, but back when Emily was "up and coming", she would have been ideal. We need a newcomer with an Emily-esque quality in that role.
Okay, I think I have my dream cast assembled:
Desiree-Michele Pawk
Fredrik-Michael Rupert
Carl Magnus-Howard McGillin
Charlotte-Judy Kuhn
Petra-Marla Schaffel
Anne-Kelli O'Hara
Henrik-Danny Gurwin
Mme. Armfeldt-Betty Buckley

Love your cast MEF...Perfect...Still bummed I didn't get to see Judy in WOMAN IN WHITE...
MEF- if only... *swoons*
Melissa Errico Fan- Michael Rupert as Frederik? Now, I do love Michael Rupert, but I think that Frederik has to have more magnetism to him. If he is a man who can attract THE Desiree Armfeldt.
And Howard McGillan is definitely not right for Carl Magnus. I would love to see someone like Kevin Kline in this role, in my heart of hearts.(he is also someone who could play Frederik) I think physically you need someone along the lines of a Burke Moses, Brent Barrett, etc. I'd be curious to see what Stokes would do with the role.
What about John Lithgow as Frederik? I seem to be in the minority when it comes to those who liked Jeremy Irons. Though he was somewhat ill at ease, I liked a lot of his choices. Now, I don't know if either of these men can sing, but what Tim Robbins or Liam Neeson as Frederik?(I've always been curious about Robbins on stage) And pairing either of them up with their wives on stage in the two leads would be fantastic, in my opinion.
Pinaud --- I love your idea of Lithgow as Frederik! He'd be a great choice.
As far as Carl-Magnus, remember he's Desiree's current "boy toy." He needs to be young (enough), virile, and great-looking... because everything else about him is annoying and pompous. Kevin Kline and even Howard McGillan are past that age and "type" now. But Kline would also make a great Frederik.
EDIT: And MB--If Marin did play Mala, this musical would turn into "The Mala Show!" No set piece would go un-chewed, and no spotlight would go un-stolen. And I mean that in the most loving and kind way.
