*Spoilers for Book of Mormon below*
When I saw The Book of Mormon in Chicago about a week ago, one moment of dialogue in act 2 got a moment of laughter that evolved into applause, specifically the scene where the other villagers tell Nabulungi how obvious it was that Arnold's stories were made up and ridiculous.
Also, when I saw Spamalot in Detroit a few years ago (during the heart of a scandal with Kwame Kilpatrick, a mayor who got into a fight with a sex worker among many other things), the Knights who say Ni reintroduce themselves in a nonsense word ending with "Kwame is in texas." Then adding that he'll probably return, as strippers fight back there. The line got so much laughter and applause that it stopped the show for a solid minute.

One off the top of my head is Megan Hilty's delivery of the "Rooster to a Hen" monologue in 9 to 5. I'll post more as/if I think of them.

I just did Godspell:2012 in Pittsburgh. The Donald Trump sketch got a lot more laughter and applause than it did five years ago, when Trump as a serious political candidate was a gag and not a bizarre reality.

I remember, in the last revival of Steel Magnolias, the line about seeing so much smoke it looks like they're electing a new Pope getting HUGE applause/laughter, because there really was a new pope being elected at the time. Sometimes the stars align!

This didn't happen in previews, but a few weeks ago I caught a matinee of Waitress and Keala Settle got applause for her comedy a few times - once just for the expression on her face.

In American Psycho Patrick Bateman got crazy applause for his line about watching a Sally Jesse Raphael show in which Trump appeared. Also when Van Patten talked about Bateman's hero The Donald in another scene.
