I know it's an unpopular opinion but I loved DRV's Fantine and came over from the UK specifically to see her (and others) in the Les Mis revival a few years ago. I'm returning to NYC next week and I know she's doing a play while I'm there. I would love to get her to sign one of my Playbills from that revival and am curious about the best way to go about it without having to drag my brother to stage door on the off chance, iykwim.
Should I send a letter and the playbill to the theatre? Does she even reply to such requests? Any other thoughts?

I once mailed a Playbill to Terri Hatcher when she was doing the national tour of Cabaret. I saw the matinee and she didn't come out the stage door. I included an envelope with stamps to cover the postage. Never got it back. Don't know if it got lost in the mail or what. Always a risk.
Now, I play it safe, and I would get the autograph in person if I can. Since you are in town, that is what I would do.
Go to the theater a bit early before the show, and leave a note at the stage door asking that it be given to DRV. This way, she will know that you are waiting for her after the show.
There is an address on her official website, but it asks for a postage-paid envelope. It is a hassle, especially for overseas mail.
