When I saw DRS over February break last year, it was full of families (a Wed mat) and literally a cell phone went off every five minutes. So finally Jon Pryce turned to the audience when one rang right after "Dirty Rotten Number and said "Well, I guess that's my agent calling."
Another from that performance was right before when Pryce says "Oh God its like the David" some girl started laughing when he slapped his. Pryce turned to her (she was in the front row I believe) and said "And you think YOURS is better?"

One time during Wicked, Glinda's bed was supposed to roll offstage, but it didn't. So when the classroom set came on, there was a bed in the classroom. William Youmans comes on, takes one look at it and goes...

at the producers in november, after Ulla's "When you got it, flaunt it" number and she asks Max(john traecy egan) and leo (hunter foster) "okey dokey, well you like it?" someone in the audience yelled "OH YOU KNOW I LIKED IT" so John Egan says "This man pays 100 bucks for a ticket and he thinks hes funnier than mel brooks"
About 5-6 years ago, Audra McDonald was scheduled to do a benefit concert at the Prince Theater in Philadelphia. McDonald took ill and the benefit was postponed.
At the concert, after she sang the first couple of songs, the latecomers were seated and, of course, there was a bunch of them sitting in the front few rows right in the middle. As they took their seats, Audra looked at them and said coolly "Glad you could make it". One of the latecomers immediately answered back "Glad YOU could make it!" (In explaining her absence the previous month, Audra said she sounded like "Bea Arthur on speed".)

This is in reference to an old post, but I thought that bit with Millie falling off the chair was a regular bit. I did a production with the original staging and we had that. I suppose it could have developed out of something that happened accidentally the first time, or perhaps one night she did a more spectacular fall than she intended. If they both cracked up, maybe that was the case.
The best "ad-lib" wasn't done on purpose. In The Wedding Singer, Robbie (who wasn't being played by Stephen Lynch that night) came out on stage with his fly unzipped and his white shirt sticking out of it! The audience was cracking up and so were the actors. It was the scene where Julia is at the Grandma's and she throws the papers at Robbie and says, "You're an asshole!" It was supposed to be serious, but no one was. He was literally the last to realize. He then looked down and pulled it up. It was classic!
