Not being a musical theatre actor, I can't imagine the effort it takes to perform some of those roles. As a dramatic actor, playing John Merrick in The Elephant Man was pretty taxing.

hmmm...i would have to say Jesus Christ in JCS. First and foremost, because of the vocal range of the show. And THEN!! the 39 LASHINGS...and they were REAL. with REAL whips. Yes, folks! I said REAL. And mind you, one of the guards was this ENORMOUS scary guy who kind of bumbled around the stage for the most part, but when he had that whip in his hand...his percision was flawless. *winces*
But definitely rewarding.

i was in schoolhouse rock, live! (yes, i give you persmission to laugh) - and it was 27 uptempo songs, all incredibly intense dance, and i was in 26 of them. it was so hard!!
mind you, we didn't have any costume changes to complicate anything, but we did have to lift these MASSIVE wooden cubes and move them around all the time in between dances. let me tell you - i was IN SHAPE after that one!
A Chorus Line. All but one of us weren't dancers, and the choreographer would make us wear our dance shoes all day, any where we went. Not to mention, he would have us run up and down stairs and on treadmills before we came in to sing our songs to condition our bodies.
My feet hurt. My legs hurt. My voice hurt. My hair hurt. I was very, very, very happy when that show was through...
A Wrangler (cowboy) in Will Rogers Follies. I'm NOT a dancer, but got roped into it -- pardon the pun -- by my friend who was the music director. They had somebody drop out and she knew I could sing it. But of course, it's a dancing role, and I worked my ass off! Still felt like a fool, though! :)
