I just saw "Oh Hello" on Netflix last night.
During the run, was Matthew Broderick always backstage?

No, he was not. It was one of the funniest additions for me as someone who saw it multiple times that he was actually behind the door. For most of the run actually that line was about F. Murray Abraham and not Broderick.
