Not sure of the exact process for joining, but I think that most Broadway ushers (the only exceptions might be Roundabout's and MTC's houses, but I'm not sure of that) are part of Local One of IATSE (International Alliance of Theatrical Stage Employees) which covers stagehands and box office personnel among others. You could contact them (or check out their website or heck, ask an usher next time you're at a Broadway theatre) for more info.
FYI -- You can volunteer usher at nearly all Off-Broadway houses, however.
