If you see Falsettos as one show, "the lesbians from next door"

I don't think Pharoah in Joseph is an important/main character

Miss Stacy in Anne of Green Gables. Bet none of you thought of that, huh?
I played that role in middle school, and every night I sat backstage and played cards and amused myself for an hour and a half before my character showed up (a few scenes into the second act). She's pretty important, as she causes Anne and Gilbert to enter whatever thing it is they enter and then admit their undying love for each other or...whatever it is they do.

The most famous example is Tartuffe.
Others include Hickey in The Iceman Cometh, Autolycus, Perdita, Florizel, Old Shepherd, and Clown in The Winter's Tale, Leslie and Sarah in Seascape, Mr. Lundie in Brigadoon, Lee Harvey Oswald in Assassins, the Starkeeper in Carousel, Big Daddy in Cat on a Hot Tin Roof, and Danforth in The Crucible.

Motormouth Maybelle in Hairspray
And Lola in Damn Yankees doesn't show up for a good 40 minutes, maybe longer.
jasonf: Freddy appears in the very first scene of MY FAIR LADY.
Not exactly standard rep but Babylove in The Grass Harp, both the play and musical.
Jane in Tarzan?
Uncle Oscar in Little Mary Sunshine doesn't come at all until Act II
O.B. Abbott in Radio Gals comes like half way through Act I

Ulla doesn't enter "The Producers" until after "Keep It Gay", and from then on, it's about two numbers then the Act One curtain.
Trix has a second during the beginning of "Drowsy", then disappears for about an hour and a half.
the Mikado in "The Mikado". He doesn't show up until like, the second half of the second act?
Enjolras doesn't show up for a good while into "Les Miserables". Same with Ellen in "Miss Saigon".
Doc in West Side Story. Though he's more of a supporting role, I suppose.
Even so, he's got an important part - especially at the end (ie lying to Tony and therefore, causing Tony to find Chino and thus concluding with Tony getting shot by Chino).
"Yes, the brutalities of progress are called revolutions. When they are over, men recognize that the human race has been harshly treated but it has moved forward." - Les Miserables

What? Are you talking about Amneris? And if so, she is in the very first scene in the show and sings the first song.
The Phantom doesn't appear on-stage for quite a while in POTO.
Charely's Aunt doesn't come on-stage until half way through ACT II of Charley's Aunt

A main character is a role like Joseph, a role like Pharoh is supporting.
Clearly the original poster meant Amonasro. Not difficult to figure out.
I can't ever understand your original post because the grammer is so bad.
