I would guess no later then 4:30. And that may still even be too late.
I was fine at 4:30 on Tuesday (7th in line), but that may change now that it's a weekend. I'd say 5 PM just to be safe.

I've decided to go for the Saturday Matinee, I don't think I have a chance anymore. I was thinking about getting there 9:45-10ish. Is that good or do you think that it is too early or too late?

Do they only give out the SRO at a certain time? Can I get them when the box office opens?
The box office starts selling them 2 hours before curtain. People start lining up about 1-2 hrs before the two hours before curtain.
Get there as early as you can if you want a ticket. Just bring a book or something.
We got there around 4:15 and were first in line on wednesday night.

I'm planning on getting there two and ahalf to three hours ahead of time. What are my chances at getting a pair of tickets?
I think it is very unpredictable because it is the first weekend after the Tonys. Also, I do not think they sell standing room until after the show is sold out.
I went to see the show last Sat night. For that Sat afternoon, the show was not sold out until around 1pm, so I do not think they sold standing room until then. In any event, there was no standing room line at the box office when the "this performance is sold out" signs went up for the matinee at 1pm; I assume that this is because it wasn't clear whether they were selling standing room at all, but I am not sure of that. It is possible they sold standing room anyway in anticipation of their selling out and they were all gone already.
For the evening, I believe that the last person to get standing room tickets showed up around 5:10pm. The Sat evening performance was sold out before the matinee was, so it was clear that they were going to sell standing room that evening.
