The location you stay in depends on what your goals are for the trip and what your budget is for a hotel. I stay in midtown in the theatre district because my main purpose is seeing shows back to back. I want to be close to the hotel to freshen up or rest. I don't really do a ton of site seeing.
If you want to stay downtown to be closer to attractions there or uptown then you need to know what you want to be doing.
I get great values midtown at The Row, The Hudson, The Apple Core Hotels.
