I saw this last year at the Magic in SF and my first though was that it had to be Off-Broadway. It’s a very intimate show that would not work well in a large venue. I thought if it was to have a Broadway run it might work in the Hayes or Booth. The Magic has only about 150 seats.
This is not a production number musical, in fact they make a joke about that when it looks like a production number is starting and Dede yells out to them to stop and says something to the effect of “It’s not that kind of f**king musical.”
Side note---“The Opposite of Sex” in it’s SF form was in much better shape then the current SF run of “The Mambo Kings”.

Opening Off Broadway is absolutely the way to go for this show. A musical based on THE OPPOSITE OF SEX would have bombed in a Broadway theatre, but Off Broadway, where they can operate at a fraction of the cost, they'll be able to have cheaper tickets which will appeal to a younger audience and thus have the potential of actually making a profit!
