See I don't think with too many of my favs there's one moment that makes me want to go back, it's a lot of things. But to answer the question slightly different for some I've seen more than once:
DRS-the fantabulous performances
Wicked-DG, along w/ Wizard and I, No Good Deed and the inspiring story
Rent-the amazing book and story, but partiuclarly when Seasons of Love, Will I?, I'll Cover You Reprise and One Song Glory are performed--though none are my absolute fav Rent songs, they're the ones I love to see in the context of the show and live
Phantom-recently Hugh Panaro's magnificent performance, but also the title song, Music of the Night, Angel of Music and All I ask Of You, combined w/ the spectacle of the smoke and lake and orchestra
Light in the Piazza-the fact that there's so much to that show-orchestra, scenery, music, performance everything, that I couldn't catch it all the first time and I got a totally diff. experience going back the second time
La Cage-watching Les Cagelles, particularly in "La Cage Aux folles"-guess that comes close to a moment
J/H - TITM & Confrontation
Civil War - The Glory
Scarlet Pimp - Into The Fire
In Les Miserables when they say the line
"To love another person is to see the face of God" or just that whole show in general considering I have seen it 9 times altogether!

* when Elphaba rises up in Defying Gravity and starts that "So if you care to find me..."
* when the chandelier rises from the stage during the Overture of THE PHANTOM OF THE OPERA
* when the candles rise from the floor during the title song in THE PHANTOM OF THE OPERA
* EVERYTHING in CHICAGO.
There's a great moment in the Stratford production of Into The Woods, so if you don't want a spoiler, don't read this:
During No One Is Alone, when Jack and the Baker first come in and the Baker reveals the death of Jack's Mother, the guy playing Jack became beautifully dramatic and began to walk off-stage to kill the Steward. Before He left though, the Baker grabbed Jack and held him back while singing his lines. This moment was so wonderfully acted, I nearly started crying.
**End Possible Spoiler**
Also, Seasons of Love is always a great song - there's so much energy in the entire theatre and from the cast. I loved it!

I agree with gavrochegirl's : When Elphie Defys Gravity (wrong spelling somehwere, don't kill me!) or in Rent durring Seasons of Love. Or Hairspray durring the fanale number You Can't Stop The Beat. Chills!!!!!!!!!!!
