The Drowsy Chaperone and Miss Hannigan immediately come to mind.
Princess Fiona hands down the funniest female role I have seen thus far. I don't care how old you are, the farting and burping song is pretty darn funny.
Also Val from A Chorus Line was pretty funny. And Debra Monk's character in Curtians [I can't remember her name.]
Four words: Delilah. Strict. Zombie Prom.

Cunegonde in CANDIDE is probably one of the most impressive female comedy roles. You have to be really, really funny and hit 21 (?) high Cs in one song. No biggie.
I like my funny ladies on the tragic side so I would have to say either of the two leads from Grey Gardens or Mrs. Lovett. All have hilarious lines and lyrics that make me laugh every time I listen to their albums.

Edit: The Adult Women in Spring Awakening has some pretty funny scenes.
Jolene - Dirty Rotten Scoundrels
Jessica Cranshaw - Curtains (even though the part is tiny)
are we talking about leading roles or character parts? because Madame Maude P. Dilly was a pretty funny part. and I agree with a lot of roles that were already posted here. Miss Adelaide is probably one of the gold standards, though.
Mazeppa, Electra, and Tessie have my vote.
