"This isn't a movie, but "Ugly Betty" does it for me.
I love the stylized look of her Queens home and the tacky NYC office. It is such a twist on the typical approach to showing off the city. "
Agreed.
I really like:

MIRACLE OF 34TH STREET {remake or original}

"Keeping The Faith" because I was an extra and you can actually see me if you look very close in two scenes in the film:
The Temple and the Church.
It was a blast!
And "When Harry Met Sally"
"You've Got Mail" because I love Hanks. Always have.
Hanks is the greatest and sweetest actor in the Universe.
I was an extra in You've Got Mail by accident and met Tom there.
He is one of the nicest people I have ever met.

Two Weeks Notice was another great one. However it wasn't an NYC shot that makes me love it. When they get dropped off in the strip mall parking lot in Jersey, that is right in my area and was a hoot to see!

Nop one has mentioned Scorsese's AFTER HOURS - a great depiction of the funky downtown lifestyle.
While we're at it - TAXI DRIVER for seedy Manhattan in the 70's, GOOD FELLAS for Queens, MOONLIGHTING for Little Italy...

When I was a wee lad I truly believed I was going to live in the East Village in a giant loft with samurai posters on the walls and a rooftop area with a hammock where I could cuddle my cat, drink vodka from the bottle, and eat Chinese take out while saxophone music drifts from another apartment. And all on a projectionists salary! Yep, its DESPERATELY SEEKING SUSAN.
"Impossible is just a big word thrown around by small men who find it easier to live in the world they've been given than to explore the power they have to change it. Impossible is not a fact. It's an opinion. Impossible is not a declaration. It's a dare. Impossible is potential. Impossible is temporary. Impossible is nothing.”
