No but you do have to join their specific union. Contact The Shubert Organization, the Nederlanders etc. to find out specifically how to apply.

IATSE, Local 306 is the union. You apply at the theater organisations office like you do at any regular job (fill out an application). However, members of the union are given more weight (you don't need to be working to join the union), and you will only work as a sub filling in for someone who took time off. I believe the Jujamcyns are the easiest to get a job with. But they tend to be strict about the placement process, so if they don't like you the chiefs won't bring you back to their theater.
The Shuberts are less strict about placement, but no one seems to get hired through the main office. Everyone I know who works for them has been brought in by the chief of a theater. And again, you will only work as a sub.
The best bet is to get the chief usher to bring you in as a sub. You'll get to skip the hiring process altogether.
Unless things have changed a great deal in the last few years, get old and grumpy as hell. That'll do it.
