There was a LOT of applause after Johanna in Sweeney. Like the person said before me, you just want to stand up and applaud before that! The "with or without his privates" line got a lot of laughs too.
A lot of Spamalot gets a LOT of laughter.
There were a few lines in Drowsy Chaperone that got a loooot of laughter. And Sutton got a lot of applause after "Show Off."
*amateur moment* When I did Beauty and the Beast at school we got a standing ovation (sold-out crowd, 1300 people) for Be Our Guest one night. It was one of the most amazing and gratifying experiences of my life.
Definitely a lot of applause after "Dirty Rotten Number," and also "GBS."
There's usually non-stop laughter during the Old lady/wheelchair dancing in "Along Came Bialy" from THE PRODUCERS.
The laughter lasted quite long after the "Leaf Coneybear has cats" line in SPELLING BEE.

When I saw Bruce Vilanch as Edns in "Hairspray," the dialogue break right before the end of "You're Timeless to Me" was probably the funniest moment I've ever seen on Broadway or any other show.
I definitely agree with all the Spelling Bee mentions! Having seen both the Broadway and Australian productions, the audience is just in hysterics for the majority of the show. A fabulous evening out!
Also, having seen both the Australian and Broadway productions of Urinetown, "Run, Freedom, Run" always bought the house down. The second time I saw it in Aus, there was a two or three minute standing ovation mid show at the completion of the number (led by yours truly with no shame at all!) The fabulous cast held their final positins for the entire ovation, only moving slightly to catch their breath and stifle their laughter. Ah - it was GREAT. But perhaps I am a little biased, as i am a true Urinetown fan.
And the other show that springs to mind immediately is the Lion King, namely the Circle of Life. Having been lucky enough to see the show here, in Aus and on the West End, the Circle of Life often receives applause from the moment the giraffes come out until Rafiki holds Simba over the heads of the animals atop Pride Rock. Pity the show goes completely down hill from that amazing moment...
Defying Gravity also often garners a lot of applause both during and at the end of the number.

Jemima, I agree with you - the break in "Timeless to me" from Hairspray is hilarious. John Pinette is excellent and I was laughing so hard I was almost in tears.

i didn't even think of wicked! haha i saw it in previews and in Defying Gravity when Idina says, "It's meeeeeeee!!!" and starts to fly everyone went crazy and even more so after the song was over.
