I have not been to the TKTS Times Square Booth since the new facility was completed. Is there generally a much shorter wait in the play-only line, and is that line easily identified?

There is a play-only window, and you don't need to wait on the line for musicals. Go up to one of the workers near the ropes at the front, tell them that you want a ticket for a non-musical show, and they'll point you to the right line.
I assume that's be the line with nobody in it?
That's handy to know!
The play only line forms on the Broadway side of the booth.

BobbyBubby, a lot of people who don't know if Phantom of the Opera and Mamma Mia! are musicals or not line up there.
I was under the impression that due to various situations like tourists using the play only line for musical tickets etc, that they ditched it altogether.
According to the TDF website, the play-only line still exists. Perhaps someone who has been there recently can report.
I am not sure either way on this one. I do remember going not that long ago and it not being there. I tried to ask for it when I wanted to go see Superior Donuts and they said that they didn't have one anymore. They also said that one of the main problems was that not only would people use that line for musicals, but that the people working the booth would sometimes (depending on who was working the play only line) sell them. Despite the fact that when it happened, they were supposed to direct the people's attention to the other lines.
The reason why I am asking and not sure is that because it seems to me that they have gone back and forth on the choice to keep it or not.
I waited on the line back in September and there was no "Play Only" line...I don't know if that has changed but I'd imagine it hasn't.
Interesting that some have found that there was not a play only line at times. I asked TDF if the line had been discontinued (or was only in place at certain times) and received the following response:
"tdf appreciates your inquiry and welcomes the opportunity to serve your
needs.
There is still a play only window. It is #12. Sometimes when it is very
busy there are two play only windows #11 and #12.

I don't know if this is how it still is, but when I worked at TKTS at the Marquis, the Play Only line was only really used when TKTS first started selling for either the matinee or evening. Once the initial line was cleared, they'd start to serve people purchasing tickets for musicals.
I feel like the general impression that I am getting is that the play only line is very hit or miss and is at the full discretion of the booth. I mean, I can see why it would be helpful. However, I can also see why they might try to minimize its usage. Like I said before, I can't tell you the number of times that someone tried to buy a ticket to a musical (hell, sometimes they were even successful in their attempt) and either they got turned away or walked away successfully.
