I'd love it if they went kind of the 'Harry Potter' route, scouting for talented unknowns as the two leads while the supporting older cast featured established actors. Meryl Streep as Morrible, as someone suggested, would be great I think. I could also see Anjelica Houston in that part, though I don't know her musical background.

I;ve always thought Anjelica Houston would be a perfect Morrible, she doesnt need a musical background since Morrible does not have to sing. Cast her opposite her ex-husband Jack Nicholson as the wizard.

I think Eden Espinosa and Megan Hilty should do it. I'm more of an Eden fan, but they work well together. No, they're not superstars. If they want to lure in an audience, hire some hunks for the male roles.
