BUT i believe we must concede that THE scene is "Puttin' on the Ritz" from the aforementioned Young Frankenstein.
The face Audrey Tautou makes when she's having sex in Amelie. Hi-larious!
When Jim Carrey takes over Steve Carell's mouth while he's anchoring the news show in Bruce Almighty.
When Will Farrell gets hit by the taxi in ELF. Never ceases to get me. Also, when he jumps onto the Christmas tree from the couch. Great stuff.
Summoning the Invisible Swordsman in THree Amigos.
"Damn! We're in a tight spot!" from O Brother Where Art Thou
Cary Grant's wild takes in Arsenic and Old Lace
The entire movies: CLUE, Groundhog Day (watching it tonight!), Waiting for Guffman, and Noises Off

I can't pick out individual scenes, but these are some movies I've seen numerous times and still crack up over:
A Mighty Wind
Elf
any of the Austin Powers movies (I know they're all the same jokes, but they still make me laugh!)
Connie and Carla
and a bunch more!

At the moment I think of scenes from:
A Mighty Wind
Legally Blonde
Misery

Here's a couple off the top of my head. I'll update as they pop up.
Best in Show - "Go and get Busy Bee! Run!!!!"
Romy and Michele - Romy whips Michele with her two-feet long hair. "Ow, that hurt. But, it looked really good."
Mean Girls - "Is butter a carb?" "Yes....?"
Never Been Kissed - Josie runs up the stairs, trying to catch up with her friends. "Kirsten, Kristen, Gibby! What's up, girlfriends?" She trips. "Ow."
Moulin Rouge - When Audrey leaves the production. "Goodbye!" Slams door. Also... "And in the end should someone die?" Pause. "So exciting the audience will blah blah blah."
Annie Hall - Annie's monologue about her narcoleptic relative.
Liar Liar - Pretty much the duration of Carrey's screentime.
Pulp Fiction - "You can get a steak here, Daddyo. Don't be a..." Mia draws a square in mid-air.
Finding Nemo - "Yeah, it runs in my family. Well... at least I think it does... Hmmm, where are they?...... Can I help you?"
Almost Famous - "Are you Mr. Miller? You have a message from Elaine, your mother. She's a handful... She freaked me out, man. Tell her to stop..."
Amelie - The orgasm montage
Adaptation - The dial tone sequence. "John, I'm very happy now. Very happy."

All of Parker Posey and Catherine O'Hara's scenes in Waiting for Guffman.
The speech concerning the AC Unit in Hands on a Hard Body (my favorite documentary).
Tammy's drug-induced delirium during a church cable broadcast in The Eyes of Tammy Faye.
Parker Posey in the pet supply shop in Best in Show.
Sissy Spacek explaining how she shot her husband in Crimes of the Heart.
Camille Coduri as she walks into the pole knocking herself unconcious in Nuns on the Run.
Jamie Lee Curtis's pole dance in True Lies.
Cameron Diaz "I can be jell-o! I have to be jell-o!" in My Best Friend's Wedding.
Uncle F**ka sequence in South Park: Bigger Longer and Uncut. Can we please have another South Park musical movie? Please?
Carey Grant face-shoves Katherine Hepburn in The Philadelphia story.
Edith Evans "A HANDBAAAAAAAAAG???" in The Importance of Being Ernest (1952).
Holly Hunter's entire performance in The Positively True Adventures of the Alleged Texas Cheerleader-Murdering Mom.

I can't help myself:
"Are you my conscience?" in Finding Nemo.
The trust-fall sequence in Mean Girls.
Drew Barrymore "You know what's a weird word? Fork!" in Never Been Kissed.
Dawn French trying to shatter glass by singing in Harry Potter and the Prisoner of Azkaban. She is a comedic genius and should be in more films. I have every episode of Vicar of Dibley and French and Saunders available on DVD and am now collecting Murder Most Horrible.

The entire "in the elephant" scene from Moulin Rouge.
And, even though it's not a comedy, when Jane mimics Blanche at the begiining of "Whatever Happened to Baby Jane?" when she asks about her fans and the reruns of her old movies ..."Do they like me?"
