The scene in "Dead Man on Campus" where Cooper takes his roommate's stethoscope and puts it down his pants, and announces "I can hear my heartbeat through my penis!" It makes me laugh everytime. I think its the residual crush I have on Mark Paul Gosalaar from his Saved by the Bell days. =)

That makes me shudder every time I think about it. I think my favorite Edith Massey moments were in Polyester.
I totally agree with In and Out and Bullets Over Broadway. Wiest also cracks me up in Hannah and Her Sisters and Parenthood.
I must also add the dinner table scenes in Eddie Murphy's Nutty Professor films. Genius!
The final scene in Some like it Hot, when Jack Lemmon finally comes clean and the immortal line "Well nobody's perfect!" comes out of his "boyfriend's" mouth. Still funny and cracks me up
(Who was his boyfriend in that movie?)
Not to mention Tony Curtis ending up with Marilyn Monroe.
Every scene in Coming to America but my absolute favorite is the scene at Lisa's House when Darryl keeps knocking at the door and John Amos's character sics the poodle on him. HILARIOUS! No wait, the nightclub scenes, especially the part where Arsenio Hall is in Drag, "I've been watching you all night and I wanna tear you apart. And your friend too." God I love that movie!
Way too many to mention, but this line usually cracks me up every time from "As Good As It Gets"
"Where do they teach you to talk like this? In some Panama City "Sailor wanna hump-hump" bar, or is it getaway day and your last shot at his whiskey? Sell crazy someplace else, we're all stocked up here. "

Just about any scene from Planes, Trains and Automobiles. John Candy had to be the funniest man ever.
I would agree with an eralier that mentioned Madaline Kahn in What's Up Doc....hilarious.
Sandy Dennis in the original The Out Of Towners with her constant lament about her weak ankles. Jack lemmon was a riot in that movie also.
Rita Moreno in "The Ritz"...I can watch that over and over...and she still makes me LMAO !!!!!!!!

GET OVER IT! mother-fing best moving in the world. martin short (aka GOD) plays this crazy ass who directs a musical version of "midsummer nights dream" at a high school. so if you call yourself a theatre person, watch this movie. some of my favorite lines:
"felix-i've taken pisses longer than your last three relationships."

All Mel Brooks movies.
All Muppet Movies (and Muppet Shows, for that matter. What can I say, I like the Muppets) except for "Muppets from Space." That one wasn't as good.
So many that have already been mentioned, to which I must add Peter Sellers in "The Pink Panther". ALL of his scenes break me up. The one where he leans against the spinning globe leaves me in tears from laughing so hard.
