Because its a song of enpowerment and its the moment where Elphaba must make a huge sacrfice and leave behind all she's known to do what she thinks is right, she leaves her best friend and her dreams behind. The first time I saw it, when Idina sang it, I cried. It was my birthday and it was so powerful.
When Shoshana and Eden sang it the times I saw it, I clapped loudly, but no crying.
I cried during Wizard and I when I saw Shoshana, because I knew all she dreamed of would never come true.

Zeitoujo...maybe because it inspries some people. Perhaps it is a tad overrated, but many people enjoy it and are inspired sometimes by its performers, etc.

Why do people cry at Defying Gravity? Because they want to. Because it's some diva belting her tits off and going really high in the air - same reason they cry during Don't Rain On My Parade...
That's assuming that people cry in DROMP, though. I bet if Idina sang DROMP then people would cry. (How much are you loving my acronym for that song?) O wait, DROMP doesn't make Fanny fly! Bugger, my theory is ruined.
I think it's probably the same reason that people give standing ovations when a show isn't that good. They've paid money to cry and to ovate... £60 in London... if they can't whip a few tears out for £60 then what is the point?

I personally don't feel WICKED is a show to cry over. It's just too corny to get my emotions.
If people cry over WICKED, how would they deal with FOLLIES? Well granted some of the crazy Wicklyn's wouldn't get FOLLIES.
