Unless I missed it, I thought Marilyn Cooper in WOMAN OF THE YEAR would be one of the very first posts. Her portrayal of a suburban housewife singing "The Grass Is Always Greener" with Lauren Bacall only lasted a few minutes but won her a well deserved featured Tony award.
Judge Turpin is actually only in one scene in the second act of Sweeney.

To add on earlier: every character in Lestat with the exception of Lestat, Armand in the ensemble are in either one act or the other.
The Phone Man in Little Voice (a role I recently played) is only in Act I, Scene I. Such a boring role after that...
I can't think of them by name, but many characters in the play Spring Storm by Tennessee Williams are only in one act. And although I only vaguely know Violet, I'm pretty sure there are several characters that don't reoccur.

Actually if the show is done as written that actor is on stage a ton, he literally plays every character seen or heard aside from Seymour, Audrey, Mushnik, the Urchins, and the Plant.

In the workshop of Dreamgirls, Effie was only in Act 1. Then Jennifer Holliday said "I'm not showing up to belt a showstopper and not be seen in Act 2." So Michael Bennett had to reconceive the show with an Act 2 Effie. But for one brief moment, there was an Act 1 Effie.
