The first character that comes to my mind is Fraulein Kost in Cabaret who has a fairly prominent role in the first act but is never seen nor mentioned in the second.

Ulla in THE PRODUCERS
Ulla is in both acts, though she appears late in act 1.
Sarah appeared in Sarah Brown Eyes in both the original and revival of Ragtime. She also appears in the finale.

Then he comes into the apartment out of breath (the joke being that they live in a 5-story walkup) and leaves, never to be seen again.

They're not really major characters, but Officer Welch and Officer Pudney don't appear until (late) Act Two of RUMORS.

The latest appearing major character ever is probably Glinda in THE WIZ. In recent productions, the actrees doubles as Aunt Em.
