Sarah comes back in act II during Sarah Brown Eyes. At least she did in the original; I never saw the revival. The first character that comes to my mind is Fraulein Kost in Cabaret who has a fairly prominent role in the first act but is never seen nor mentioned in the second.

Since you didn't specify plays vs. musicals, I'll throw BAREFOOT IN THE PARK into the hat. The deliverman literally has one line: "Bloomingdale's!" Then he comes into the apartment out of breath (the joke being that they live in a 5-story walkup) and leaves, never to be seen again.

Really Jimmy and Muzzy come in late in Act 1 of Millie. Jimmy has that brief street scene after the opening number and reappears very close to the end of Act 1. I remember chilling quite a bit backstage during Act 1
