For me it's usually the split second as the lights fade up after the set change between the third and fourth scenes.

It was probably when that arrogant asshole Thespis started doing callbacks in the middle of a dramatic religious chant ritual in ancient Greece.

They probably killed him, but now we have SHREK.

Didn't theater's singular defining moment actually occur on TV, when Cheno and Esparza appeared together on Pushing Daisies?

No, it was when Cheno and Idina both appeared on GLEE, thus creating a theatrical vortex in which the two will have no choice but to return to WICKED together and finish out the run of the show.
