The Lion King- the scene where the actors dressed as the animals walk down the aisle(if I remember correctly I think that's in The Circle of Life).
The Producers- When Leo and Max first meet and Leo talks about his blanket. That scene's so funny!
Beauty and the Beast- The transformation and the end of the show(the music at the end gives me chills)
Footloose(even though I haven't seen Footloose and was just in the show I can still name a scene I wasn't in, but saw)- Holding out For A Hero and any of the other scenes with Rusty, Urleen, and Wendy Jo.

The last scene of the first act of Sweeney Todd ("Pretty Women," "Epiphiny," and "A Little Priest")
Though I believe it includes up to 3 to 4 scenes, I'll say the entire sequence from Ragtime of "Your Daddy's Son" though "Wheels of a Dream."
The reprise of "Waitin' For the Light to Shine" from Big River, especially in the revival.
"Forever Yours" from Once on this Island.
"As Long As He Needs Me" from Oliver!
"The man who wishes he had written Phantom of the Opera!" - SueleenGay

In no particular order (random from my brain)...
Act 1 (Little Priest) scene in Sweeney Todd.
The bedroom scene (Popular) in Wicked
The horse race scene (Ascott Gavotte) in My Fair Lady
The New York bar scene toward the end of Brigadoon
The opening audition scene from A Chorus Line
The flying off to Neverland scene in Peter Pan
The scene where Rosabella teaches Tony a little English in Most Happy Fella
The elevator scene from Sweet Charity
Desiree's bedroom scene (Send In the Clowns) in A Little Night Music
And just to show it ain't all about musicals...
The "Marry me, Howard" scene from Picnic
The stigmata scene from Agnes of God
The first Maggie and Brick scene in Cat on a Hot Tin Roof
The gentleman caller scene from Glass Menagerie
The final scene in The Heiress when Catherine climbs the stairs
Oh, and I'm still cracking up over "Norbert Leo's Butt" (It's the juvie in me!)

In no order:
MUSICALS
1. When the stage explodes with light, right as the opening chords of "Rent" start.
2. Thurio's Samba in 2 Gents (with the dancers flipping him off while dancing)
3. I'm Not Dead Yet
4. It Sucks To Be Me
5. Springtime for Hitler
PLAYS
1. When Michelle Lee made out with Valerie Harper, and then Tony Roberts a few minutes later in Allergist's Wife
2. The "pee-in-your-pants" jolt in Pillowman
3. The final scene of Doubt, where Sister Aloysious confesses to Sister James that she lied to Father Flynn. This is perhaps my favorite scene of all time.
4. "All the World's A Stage" in As You Like It
5. The final scene of Bklyn Boy where Adam Arkin recites the mourners kaddish.

I love love love the scene in Wicked where Elphaba and Glinda fight eachother in Act II. Shoshana and Megan do it wonderfully, and it gets the audience rolling everytime.
In Hairspray, and I don't even know if you would consider this a scene, but after curtain call, when Tracy says, "Everybody! Let's Dance!" is awesome. When I saw the show, audience members were dancing in the isles and everything, it was a fantastic end to a fantastic show!
