I've seen way too many shows since the 1950s, to limit it to one. Although the best musical I ever saw was the original London production of My Fair Lady, with Julie Andrews and Alec Clunes. But that wasn't on Broadway.
On Broadway, there are just so many. Seesaw certainly remains the best danced show I ever saw, and Ragtime restored my faith in the American musical. As for non-musicals, the original The Miracle Worker and Wait Until Dark, are favorites, among many.
