There's a bunch and I'll come back to this thread when I can actually think about it, but off the top of my head, there's a line from THE HOURS that I actually memorized when I was 17, I believe which was when I saw the movie in the theatres.
Meryl Streep's Clarissa is having this really awkward, sort of painful moment with her best friend Richard (played wonderfully by Ed Harris). He tells her, "sometimes I feel I'm only staying alive to please you," or something to that effect. And then she just stares at him and tells him "that is what we do, that is what people do...they stay alive for each other." There is so little hope in that line, because he is completely right, he makes her alive and yet he keeps her from living, and there is such co-dependency but they did share this beautiful moment way back...I don't know, but from the second Meryl uttered the line, it ripped right through me.

This is more a question of delivery than of the actual line, but in the Kenneth Branagh HAMLET, when Julie Christie (as Gertrude) said, "Oh, Hamlet, thou hast cleft my heart in twain"--I believed it. It's one of the few moments from that movie (most of the rest also involve Christie) that still cut through me when I think about them.
I ask in all honesty/What would life be?/Without a song and a dance, what are we?/So I say "Thank you for the music/For giving it to me."

The line is the last one in this scene. The way it's delivered, and the context, just rips my heart out.
SCHINDLER'S LIST
The scene:
Oskar Schindler: I could have got more out. I could have got more. I don't know. If I'd just... I could have got more.
Itzhak Stern: Oskar, there are eleven hundred people who are alive because of you. Look at them.
Oskar Schindler: If I'd made more money... I threw away so much money. You have no idea. If I'd just...
Itzhak Stern: There will be generations because of what you did.
Oskar Schindler: I didn't do enough!
Itzhak Stern: You did so much.
[Schindler looks at his car]
Oskar Schindler: This car. Goeth would have bought this car. Why did I keep the car? Ten people right there. Ten people. Ten more people.
[removing Nazi pin from lapel]
Oskar Schindler: This pin. Two people. This is gold. Two more people. He would have given me two for it, at least one. One more person. A person, Stern. For this.
[sobbing]
Oskar Schindler: I could have gotten one more person... and I didn't! And I... I didn't!

This may sound silly, but I love the scene in SIXTEEN CANDLES between Paul Gleason and Molly Ringwald.
Something about the way she says--talking about Jake Ryan--saying "All I do is just follow him around like some kind of puppy. It just hurts" brings back adolescent longing all over again for me.

I don't think anyone who ever saw the early eighties film TESTAMENT ever quite forgot Jane Alexander's "Who did this? God damn you!" moment.

In TESTAMENT, I remember Jane Alexander's sudden frantic search throughout the house for a specific object more clearly (to say more would be to spoil one of the most powerful moments in the film). Really heartbreaking.
It isn't a line of dialogue, but there's a moment in MASH where Bud Cort is told by Robert Duvall that he is responsible for the death of a soldier. Cort's stunned reaction and slow dissolve into tears is the most powerful thing in the entire movie.

Oh, Robbie, mine is similar to yours:
"Neeeeeeeeeeeeeeelllyyyyyyyyyy"
Seriously though, the scene in Mockingbird (no, not "Hey, Boo" although that is a classic) when Atticus is putting Scout to bed and she asks to see his watch. She says, "Jem says this watch will be his one day," and Atticus says, yes, that it is customary for a father to pass his watch on to his son, and Scout asks, "Atticus, what are you going to give me?"
That simple question, for some reason, knowing what is to come and what Atticus is actually going to "give" her gets me every single time. It is such an innocent question, yet so full of meaning in context of the story.

I'm nothing if not a cliche for loving this exchange:
"It's not your fault."
"Yeah, I know."
"It's not your fault."
"I know"
etc.
That is one of the most heartwretching scenes I've ever seen.

My favorite Holly Hunter moments:
In RAISING ARIZONA, the heartbreak with which she says, "My fiance left me" grabs me every time, not least because of the way she mispronounces "fiance."
And of course: "You give me that baby, you warthog from hell!"
