"Anything You Can Do I Can Do Better" from ANNIE GET YOUR GUN
It can be "killer" based on what you do with it, just as most material goes.

"The Spectacle has, indeed, an emotional attraction of its own, but, of all the parts, it is the least artistic, and connected least with the art of poetry. For the power of Tragedy, we may be sure, is felt even apart from representation and actors. Besides, the production of spectacular effects depends more on the art of the stage machinist than on that of the poet."

"Friendship" from Anything Goes is hilarious, again, as CapnHook mentioned, if you have the right people.

I would look at the many, many parodies done by Forbidden Broadway. Also, there are a few in The Drowsy Chaperone.

I would definitely say The Song That Goes Like This from Spamalot or Any duet from The Drowsy Chaperone!

This isn't technically Broadway but "I'll Never Tell" from the Buffy the Vampire Slayer musical episode (entitled "Once More With Feeling") is a hilarious duet.

You're reminding me of people you hear at the movies asking questions every ten seconds, "Who is that? Why is that guy walking down the street? Who's that lady coming up to him? Uh-oh, why did that car go by? Why is it so dark in this theater?" - FindingNamo on strummergirl

"They say that just being relaxed is the most important thing [in acting]. I take that to another level, I think kinda like yawning and...like being partially asleep onstage is also good, but whatever." - Sherie Rene Scott

I've performed "The Song That Goes Like This" with a girl friend of mine in cabaret - it was always the audience favourite.

If you two know how to tap dance, then I suggest "You Mustn't Be Discouraged" from Fade Out/Fade In. It can be a shopstopper if done correctly.
Marie: Don't be in such a hurry about that pretty little chippy in Frisco.
Tony: Eh, she's a no chip!

I know it's more of a romantic duet than a comedic duet, but "Timeless to Me" from Hairspray has plenty of charming lyrics.
"Nothing is Too Wonderful to Be True" from Dirty Rotten Scoundrels
I don't care for the song that much, but "Come Out of the Dumpster" from The Wedding Singer is an option.
"Barcelona" from Company has some funny parts.
"Schadenfreude" or "Mix Tape" from Avenue Q.

Definitely "You Mustn't Be Discouraged". Even if you can't tap dance. The Comden and Green lyrics are funny enough.
Their "Carried Away" from "On the Town" is good too, if you can ham it up operatically.
Another good one is "Shakespeare Lied" from Elmer Bernstein and Carolyn Leigh's "How Now, Dow Jones". It's done as a duet on the album, but it's written as a solo in the vocal selections.

I've always had a fondness for I've Got it All from On the Twentieth Century. Follow Your Heart from Urinetown is hilarious when performed well. Also, No One'll Even Love You from Goldilocks.

Mister Matt- You are so right about "I've Got It All." That song, as well as the entire score, is hilariously funny. Probably one of the most comedic cast recordings out there.
