i would have to say wicked. before you start commenting, her me out:
yes, i know that there have been 'better' g(a)lindas, fiyeros and elphabas, but none of them had the same chemistry, ease, and vocal blend as these three. try listening to the original cast recording of defying gravity and you'll see what i mean. their voices blend so well, it's incredible.
I think In The Heights was pretty perfect (and I'm saying this because on opening night, Carlos Gomez's voice wasn't shot ). That cast brings so much electricity to the show.
I totally agree that the original Nine is pretty perfect, but I'm partial to the original cast of 1776. William Daniels, the divine Virginia Vestoff, Howard Da Silva, Ken Howard, Ron Holgate AND Betty Buckley! And the best part is that most of them are in the pretty fantastic film version.
But let's pause for a moment to reflect on the 1954 production of the Threepenny Opera, which starred Scott Merrill, Lotte Lenya, Bea Arthur, Jo Sullivan, Charlotte Rae and even John Astin. I wish I could go back in time to see it...

the Boy From Oz! Hugh Jackman, Stephanie J. Block (bway debut), Isabel Keating, Jarrod Emick, Michael Mulherin, Beth Fowler.. flawless cast. They were all stunning.
I have to strongly disagree with those who said Kiss of the Spiderwoman had the perfect opening cast.. I liked the replacement cast better with Vanessa Williams, Brian (stokes, he hadnt added Stokes yet.) Mitchell, and Howard McGillin.. and apparantly someone agrees with me because they recorded a new cast recording with that cast.. I don't agree about Light in the Piazza either.. I LOVE the show, but Stephen Pasquale was much better when the show made its premiere, and Aaron Lazar was better as replacement than Matthew Morrison, as much as I love him in other things, he was not right for the role.. forgetting that he can't pass for Italian, his voice wasn't appropriate.. Let's note the change in key they made for "il mondo era vuoto" they had made for him.
First thought was In The Heights with their Off-Broadway cast at 37 Arts. The only difference (not counting the cast additions they made when bringing the show to Broadway) was the one that made all the difference - John Herrera. He never had a weak voice and I felt that he embodied that part as completely as Lin, Mandy and the others did/do in theirs. He terrified me during the arguments with Priscilla and Mandy, brought me to tears during "Inutil," and made me want to hug him like a teddy bear at times as well. Plus, those original ensemble members and swings were (are) so perfect and has such amazing energy.
I disagree with the comments about Matt/Aaron in Piazza. I preferred David Burnham, and I also thought Patti Cohenour was at least Vicki Clark's equal in that role.
Others that came to mind: Billy Elliot! But including all 3 original Billy boys.

All Shook Up - it was a favorite of mine (you can all make fun), but Cheyenne Jackson and Jenn Gamby were fantastic and perfect for the roles, plus Mark Price, Nikki James, Curtis Holbrook and the talented ensemble were so perfect.

No subsequent productions have topped these, IMO. There are others that have equalled (Chicago) or, occasionally, surpassed (Assassins) the original, but these are the ones that remain the standard by which all subsequent versions must be compared.
