I was actually just thinking about this today while listening to Les Miz. That said...
-Jean Valjean in LES MIZ
-Judas in JCS (would love to sing "Heaven on Their Minds")
-The Leading Player in PIPPIN (Technically this could be a female role, but it's not usually. I recently got a callback for this, but lost the role to a guy - he was great by the way - so until I get it, it's on my list!)
-Riff in WEST SIDE STORY
-The Emcee in CABARET
-Seaweed in HAIRSPRAY
-Mr. Mistoffoles in CATS

HeyMrMusic - good choice. Anita was my favorite role ever!
Magdelene - I've actually seen Pseudolus played by a woman. It took me a few minutes to get used to it, but she worked it and it worked somehow...don't know how, but it did.

Don't give me too much credit. I was the understudy for Anita (I was really supposed to be Rosalia). But the regular Anita hurt her leg and I took the remainder of the run...but it was still my favorite role ever... :)

To everyone who said Judas would be the role they would want to play...in my high school's production in 2000 a girl played Judas (this was also the production where Mary Magdalene was dressed in a black patent leather dress most of the time).
For me, however, the roles would be:
