My all time favorite is "Walk Him Up the Stairs" from PURLIE.  Pippin and A Chorus Line's opening numbers are my other favorites.

Most of mine have already been mentioned, but one that I found extremely effective and memorable was Oh the Thinks You Can Think from Seussical. The Broadway production, not the WTF national tour opening that looked like the citizens of THX 1138.  It was so joyous and catchy, I couldn't get the song out of my head after I left the theatre, a couple of months before the recording was released.

I never got to see the show but played that song a lot when I got the OBCR.

Ahrens & Flaherty really nailed that Seussical opener, along with Anastasia (which I just mentioned) and Ragtime.

Agreed re: Pippin and Wicked, and I don't even like Stephen Schwartz's work much.

I wish I liked Anastasia, but it (both the film and the stage production) just bores me to tears.  I really love a few of the songs, but everything else just seems so bland and average.  I was hoping the stage version would strengthen the material and add more visual interest, but like Newsies, it had the complete opposite effect on me.  The show might do okay on Broadway during an interim in order to fill the void left by Matilda before Frozen opens, but unless it gets a massive overhaul, I don't expect a long life for the show.

...okay? I was strictly talking about Rumor in St Petersburg. Because it's the real opening number (minus the prologue)

I've always enjoyed the opening to The Black Rider, with the circus music and all the dancing weirdos coming out of the box.
