I'm not a laugh-out-loud type of person, but Avenue Q had several moments that had me doing so.
But I think Forum still holds that title with me. There's a reason it's been revived so many times.

Broadway--"noises off" comes to mind.
Off-B--without question, the old "Theatre of the Ridiculous" gang. I laughed until I cried so many times. God, I miss them.

I nearly pissed myself the first time I saw Spelling Bee.
And the lyrics for Dirty Rotten Scoundrels have to be some of the funniest I have ever heard.

Easily The Producers. Spamalot was funny, but took most of its best bits straight from the producers.
