When I saw RENT - exactly 12 years ago, the night after the Tony nominations were announced, the cast received a standing ovation when they walked out on stage to start the second act.
I've seen it happen at Jersey Boys.
And though this doesn't count: years ago at The Hollywood Bowl I believe it was Patti LuPone, Rebecca Luker and Davis Gaines in concert. The Bowl was very packed, and during Act 1, DG came out to sing his first song, Ol' Man River. Afterwards, it was a thrill to see everyone standing on their feet and clapping madly. The roar was just amazing. It must have lasted at least 5 minutes. I always think of that night because I have never experienced anything like it again.
thanks cartwheel, I was blessed to see the show at a point in my life where I really benefited from seeing it. The Color Purple wasn't the best show I've seen, but its one that has meant the most to me, and one of two Broadway shows that has ever made me cry.

Angela Lansbury after Rose's Turn in 1974 (everytime I saw it).
I wonder if the other Roses (on Broadway) got the standing O - Ethel Merman, Tyne Daly? Anyone?
Last performances should be a separate category - although I saw one of the last Side Show performances; the ensemble got a standing O at the top , just coming out and standing in those bleachers. Took me by surprise (although it was a "cult" audience, not a regular, average night.)
Raul's last "Being Alive" is the only one I have witnessed.
The reason why Rose's Turn gets a standing O, in addition to being a great, powerful song, is that many people think it's the end of the show and don't know that there's one more scene. The one I went to a few months ago had at least 30 (mostly old) people stand up and rush out the exit during the applause for Rose's Turn, thinking it was the end of the show.
At Nikki Snelson's last performance in Legally Blonde, she received a standing ovation after "Whipped Into Shape". It was started by some fans in the front row, and then other people joined in. :) It was pretty awesome.
And it was amazing
It lasted long enough that the conductor had to signal for the next line to stop the cheering.

Ethel Merman in Gypsy there were standing ovations almost for every one of her songs, but Everythings Coming Up Roses and Rose's Turn had the most people stand up for an ovation.
The most intense (and longest-lasting show-stopping) one was at the final Broadway public performance of CATS (an Actors Fund benefit)following the Jellicle Ball sequence - it wasn't a few people that started it - the ENTIRE audience literally leapt to their feet with a roar. Definitely one of my most memorable moments as an audience mnember.

I wasn't alive at the time but I have seen a video of Who's That Woman where the music hadn't even hit the final button a people were leaping (literally) to their feet.
In the 90's when Lea performed Eponine in Les Miserables. She received a standing ovation after singing "I Dreamed A Dream."
