A lot of mine have already been mentioned- Ripley in next to normal, LuPone in Gypsy, Audra in Porgy and Bess, Amy Morton in August...but to that list I would add...
Christine Ebersole in Grey Gardens (which I think is the greatest stage performance I've ever seen)
Joe Mantello in The Normal Heart
Brian Stokes Mitchell in the original Ragtime
Brian Dennehy in Death of a Salesman
I would also add one that might be unpopular...Nathan Lane in The Producers. To me, that remains the finest example of musical comedy acting that I've ever seen.

The image of Ebersole as Little Edie standing with her packed suitcase as Mary Louise Wilson's Big Edie was screaming her name is one of the most striking theatre memories. So yes, I'd definitely add Ebersole to the list, she was incredible.

One performance that hasn't been mentioned that remains indelible in my memory is Maggie Smith in Private Lives. I can't begin to describe how masterful, brilliant and hysterically funny she was. And a lesson in comic timing.

My most memorable would have to be Samantha Soul in one of Lucy Thurber's Hill Town Plays in Killers and Other Family! That's a performance I won't soon forget, she was so fearless and breathtaking in that role!!

Celia Keenan-Bolger, The 25th Annual Putnam County Spelling Bee. I saw that show many times while it was running. Every time Celia was on, she stole the show. I cried many times during The "I Love You" Song and always hoped that this would be the time when Olive came out on top.

I thought Angela Landbury's teenytiny role in The Best Man was an example of an actor spinning gold out of straw. She was so funny.
