Few things actually make me laugh while at the theatre, even the classics like FUNNY THING.. and ON THE 20TH CENTURY.
But the one show I remember ACTUALLY laughing with while watching was THE FULL MONTY. It really has an exceptionally funny McNally book.

The first time I saw Dirty Rotten Scoundrels I was laughing so much I got light headed. For me, that show takes the cake.

The most I've ever laughed at a show was Spelling Bee. It was the best hour and a half of my life. It put me in such a great mood. It was so much fun!

Mamma Mia!
I thought Hairspray was also really hilarious. I love it when Link is singing "It Takes Two" and theres that crazy girl hugging his leg LOL. Also the dodgeball part was really funny too! "Hear comes special ed. Snicker, snicker, sneer sneer." Really funny!

All kidding aside, I'd have to say Spamalot or Avenue Q of late, but if I thought about it harder I might think of some older shows that were particularly funny.

I think HOW TO SUCCEED IN BUSINESS WITHOUT REALLY TRYING has the potential to get a laugh, on almost every line. Unfortunately, the revial, blew it, in that regard.

City of Angels is the funniest musical I've ever seen, but I'll give an honorable mention to Avenue Q.
