The Producers was packed with non-stop one liners that had me falling into the aisles. Spamalot was hysterical the first time you hear it but it isn't any kind of full-blown show. Dirty Rotten Scoundrels fills that bill beautifully. It's incredibly funny - over and over again. "Chaperone" runs a real close second.

Urinetown for me. With Drowsy Chaperone now coming in a close second. I still had tears of laughter rolling down my cheeks as I was leaving the theatre after seeing both of these shows!

This last one may cause trouble but-- ALL SHOOK UP, I thought the show was hysterical, all the comedy in it, like Mark Price doing the pelvis dance during Teddy Bear/Hound Dog or when Leah Hocking's character says "Quote Shakespeare and peel me like a banana". I think its very funny. But thats just me.

I've laughed hardest at PRODUCERS, URINETOWN, SPAMALOT, and a little Off-Broadway musical called WHEN PIGS FLY.
There are some brilliant moments of physical comedy in ME AND MY GIRL and CRAZY FOR YOU - if you have the right performers doing them.

Wow. I completly forgot about All Shook Up. I had a smile on my face the entire time. I had tears of laughter streaming down my face everytime "One Night With You" broke out.

Glad to see Spamalot getting A LOT of honourable mentions in the 'Funniest Song' and 'Funniest Musical' threads on this board - esp after all the stick it's got over awards etc.
For me, it's the fact that it made me laugh till I cried (and can still bring a smile to my face when listening to the soundtrack) that is most important. I remember waiting in the lobby to pick up tickets last September and remarking to my friend (both of us veterans of numerous West End shows) that I had never heard an audience having so much fun in my whole life. They were in complete hysterics and, a couple of days later, I could understand why! Not to take anything away from any other shows (DRS & The Producers were also fabulous), but Spamalot remains the funniest show I have ever seen.
* And, just for the record, I wasn't very familiar with the Python films and have never counted myself as a particular fan of them - and it didn't make a blind bit of difference to my enjoyment!

I'm skimming through this post, and comparing it with "The Funniest Songs" thread. It is intersting to note how a few select shows are mentioned here, but can't boost a top-drawer comedy number. I'm actually checking the shows mentioned against cast albums I have to ensure I've heard the best of Broadway laughs.
...And so, there I was at the stage door for "Equus". The enthused avalanche of "Harry Potter" fans was literally pushing me into the barricades. As I was thrust face-first into Daniel Radcliffe's hat, I suddenly felt the thumb of a lanky gentleman behind me pressing firmly into my back. It was then that I realized that both of his hands had Playbills, and that both arms were outstretched in front of me...

Drowsy was hilarious (and I know it's not a musical, but I've never laughed so much at anything as I did at Inishmore!)

Two novelty off-Broadway shows I remember laughing hard at throughout were "Slut" and "The Great American Trailer Park Musical".
For Broadway musicals, I'd say Spelling Bee, DRS, and Avenue Q.

Of the Broadway shows that I've seen, Dirty Rotten Scoundrels when Norbert and Jonathan were in was the most hilarious. The whole audience was laughing hysterically the whole time.
