I remember my brother and I laughing so hard at FALSETTOS that we had tears falling down our cheeks during the first act of that show.
I give second and third places to SPELLING BEE and CITY OF ANGELS.

Definitely THE PRODUCERS. It is side-splittingly funny. I've seen it a couple of times, plus the film a few times, and I just keep laughing and laughing.

While Dirty Rotten Scoundrels may not be the funniest musical I've ever seen (though it's up there), it stands on its own as the one show I can go back to over and over and each time I laugh just as much as I did the first time I saw it.
Other funny ones are Avenue Q, Spelling Bee, Forbidden Broadway: SVU, Producers, Musical of Musicals: The Musical, and The Audience.

"If there was a Mount Rushmore for Broadway scores, "West Side Story" would be front and center. It snaps, it crackles it pops! It surges with a roar, its energy and sheer life undiminished by the years" - NYPost reviewer Elisabeth Vincentelli

I guess I'm the only one that thinks My Fair Lady is funny? The Julie Andrews Cockney gets me every time.

Definitely Spamalot and DRS, Hairspray was hilarious too.
If Percy Blakeney were in Les Mis....
Percy: Sink me! If it isn't Javvurt!
Javert: Zsah-vair, it's pronounced Zsah-vair.
Pecry: But it's spelled J-A-V-E-R-T Javvurt.
Javert: Repeat after me Zsah...Zsah....
Percy: Oh! Zsa-Zsa! Like the Gabor sister! Well I personally have always prefered Eva.
Javert: (Looks for gun)

Nunsense.
I was laughing so hard I was crying. That would have been a better straight play, really. The music isn't memorable at all, but it's so funny!!!

My favorite:
1. Forbidden Broadway- Because it makes fun of all I love and adore. And Michael West is back. I have to see the new version.
2. Avenue Q
3. Hairspray
4. Spamalot
5. The Producers
6. Spelling Bee (And now that Foa is joining the cast, I might be changing lotteries) HEH.

The Great American Trailor Park Musical
"People that excel in the arts understand that the journey is the reward...the result an added bonus. Every day I act or train is a blessing and a dream come true. If Broadway beckons so be it. I have a personal definition of success that is unshakable by a possibly unobtainable goal." -HamletWasBipolar

Lesser known? Last year, my school performed a production of "Sugar" (edited of course) during February. I can't figure out why this musical version of "Some Like it Hot", starring Marilyn Monroe, Tony Curtis, and Jack Lemmon, wasn't a hit on Broadway. I played cross-dressing Jerry/Daphne. And nothing flooded my heart with bliss like the rollicking cascading waterfalls of uproarious laughter rushing from the aisles. I honestly don't think, at this point, that a funnier show is available in a school setting.
...And so, there I was at the stage door for "Equus". The enthused avalanche of "Harry Potter" fans was literally pushing me into the barricades. As I was thrust face-first into Daniel Radcliffe's hat, I suddenly felt the thumb of a lanky gentleman behind me pressing firmly into my back. It was then that I realized that both of his hands had Playbills, and that both arms were outstretched in front of me...

Without question, A Funny Thing Happened on the Way to the Forum, with distant compatition from The Producers, How to Succued in Business Without Really Trying, Spamalot, and a good production of Guys & Dolls.
As for lesser known shows, The Mystery of Edwin Drood is quite funny, and very fun to do AND watch.
EDIT: And A Chorus Line can be pretty funny, too.
