A Day in Hollywod / A Night in the Ukraine comes to mind immediately. Little Shop of Horrors I could have sworn was originally set in LA but was changed for the film version.
For about fifteen minutes, Merrily We Roll Along.

The source movie may have been set in LA, but the show opens with a number contrasting "Downtown" with "Uptown".
LA does indeed have a downtown with its own skid row, but it has no "Uptown". It spreads out in all directions from Downtown.
I've always imagined it taking place in some northeastern or northern Midwest city.
