I think it has to be a Jerome Kern melody, since Rodgers and Berlin and Porter and Gershwin all said that he was the first American composer to inject a sense of beauty into the American musical.
So I'll pick Jerome Kern and Oscar Hammerstein's "All the Things You Are" from Very Warm for May. Many people consider it (seriously!) not only the most beautiful song written for the American musical theater, but also the most beautiful song ever written. (Period!)

To the poster that mentioned "How Could I Ever Know" from Secret Garden, I'd up the ante and say about 85 percent of that cast album would be "most beautiful" to me. I would also like to throw in "The Proposal/The Night Was Alive" from Titanic. That song is as lush as the Hanging Gardens of Babylon. Perfect.

I love this topic as Broadway shows have the most beautiful songs. On of my favorites was never mentioned, "This Is the Moment" from Jekyl and Hyde; I get goosebumps when I hear it performed. Also forgotten is the upbeat "The Best of Times" from La Cage. My next favorite is, "What I Did for Love" from A Chorus Line. I am sadly unfamiliar with Les Mis which seems to have many favorites. I also love: "You Are Beautiful/Love Look Away": Flower Drum Song, "A Wonderful Guy/Bali Hai" from South Pacific, "If Ever I Would Leave You": Camelot, "Sunrise,Sunset": Fiddler, "Neverland": Peter Pan, "Memory":Cats, "As Long As He Needs Me":Oliver, "Heather on the Hill: Brigadoon, "Make Believe" Showboat, and "Practically Perfect" from Mary Poppins. Surprisingly, nobody chose tunes from some of Broadway's longest runners like "Cats" and "Chicago". Lots of other beauties as well, but these are my favorites.
