When Mrs. Lovett is showing Toby the grinders and she shines her flashlight out into the audience during the Sweeney revival.
The eyes and mouths of the Angel statue lighting up during the “All I Ask Of You Reprise” in Phantom of the Opera.
In the “Chicago” revival when members of the ensemble sit motionless on chairs at the edge of the stage. Also, whenever Roxie or Velma come swinging out on the ladders.
Finally, during “One Day More” in Les Mis when Enjolras waves the flag and Gavroche puts up his arms.
Although it's been mentioned and then repeated, Enroljas on the barricade is my top choice. It's that single spotlight on him and the swell in the music that hit at the perfect time. And since I had been crying after "Bring Him Home" the reprise of it got me again.
The last moment in Phantom always affects me too.
I Am My Own Wife has some beautiful images in it as well (great lighting and great set!)
And I saw an unbelievable production of Titus Andronicus where the director used red fans to represent the blood at the end... It's still one of the most beautiful moments I've ever seen on stage.
Mine is the act one finale of "Wicked". The music and emotions all run through through that awesome song! It is so beautiful.
Go away Jacob.
