A VERY close runner up is actually a sound, not a quote: Little Edie's laugh in act two in response to Edith's comment "Never had a fight in my life". (priceless! and hard to duplicate-I've tried!)

One that comes to mind, is from Crazy For You, a great show, full of witty and funny dialogue.
