Barrie Ingham and George Merritt did the entire run of the original Jekyll & Hyde (along with plenty of ensemble members, some of whom did the pre-B'way tour as well).
Elizabeth Ward Land was Marie (later Marie Tussaud) in every version of The Scarlet Pimpernel.. 3 years on Broadway and over a year on tour.
On the broadway.com vlog fly girl there was an obc ensemble member who played elphies mom. No sure if shes has stayed continuously or is still in the show though.

^ If you're talking about Christy Chandler who originated the role, she is no longer there, and she wasn't consistently the Witch's Mother (also played Nessa).
Jersey Boys still has 3 original cast members: Sara Schmidt (who I believe has been there the entire time as Francine), Peter Gregus and Mark Lotito
It's not Broadway, but Jemma Rix has been playing Elphaba around Australasia since 2008 (she did start out as the standby though, but did get to perform a lot) and before that she did a shortened version of the show at Universal in Japan. She has done over 1000 performances in the role.
Ryan Sheppard has also been in the same company since the beginning, originally as Chistery and now as the dance captain.
Jemma Rix has definitely been in the role of Elphaba since 2008 and has done over 1000 shows [and is now playing in Brisbane]. She also has an amazing voice! A great Elphaba
