no, the characters are actors portraying roles.
many people are listing principlals sometimes appearing in the ensemble but OP asked for two or more designated characters. different things (Patti LuPone as Fantine comes to mind... she felt that she shouldn't have to appear in the ensemble after she dies so she conveniently missed all but one of those rehearsals)
Several actors in Batboy are playing more than one role (Sheriff/Delia, Rick/Lorraine/Mr. Dillon, Ron/Maggie/Clem, Ruthie/Ned, Mrs Taylor/Rev Billy Hightower/Institute Man, Daisy/Bud/Pan/Doctor)
Also, Spelling Bee has some of them doubling as well (Rona Lisa doubles as Olive's mother, Mitch doubles as both Olive's father and one of Logainne's fathers, Leaf doubles as Logainne's other father).

Another one that's come to my mind is Hedwig, who "acts" the characters in her life throughout the show, but then indeed transforms into Tommy Gnosis for the "Wicked Little Town" reprise.
I'm not sure if this is always the case, but when I saw Evil Dead the Musical two performers had duel roles : Shelly/Annie and Ed/"Moose".
Thought of another one. In Reefer Madness (LA/Broadway), Robert Torti played both Jack and Jesus. This wasn't the case in the movie, but the movie actually had more role splitting. In the movie Alan Cumming played multiple roles (Lecturer, Goat Man, Roosevelt, etc), Steven Weber was Jack/George Washington, John Kasir was Ralph/Uncle Sam, and Amy Spanger was Sally/Lady Liberty.

How did we forget the 1939 film of The Wizard of Oz, which double casts five roles: Professor Marvel/The Wizard; Miss Gulch/The Wicked Witch; The Strawman, Tinman and Lion/the Kansas farmhands. There have been at least three different stage versions of the film, the 1940 MUNY adaptation, a version of which is still licensed; the RSC production from 1987, which is also still licensed; and ALW's recent version, which takes many liberties with the film. Do any of them also double cast any of these roles?

Other than Youth, the Narrator, and Mother, all the characters in Passing Strange are, I think, at least tripled.
