Well, RENT was what got me into Broadway in the first place... since then, I've come to enjoy Sweeney Todd quite a bit.
Bombay Dreams and Rent.
Dirty Rotten Scoundrels or A Chorus Line.

Jersey Boys ....100%
I also love Andrew Lippas "John & Jen"
I loved Nine but Evita is my favorite. Hope they bring it back soon.
Gypsy with Into The Woods at a close second.
I ADORE West Side Story!
Cabaret would probably be a close second.
The funniest is "Guys and Dolls."
The best is "Gypsy."
But for me, my favorite is "On Your Toes." I was stuck in traffic on the 101 freeway in Los Angeles when a show about Broadway musicals on NPR began playing "Slaughter on 10th Avenue" which I vaguely remembered from having seen the revival in 1986. It just touched my soul (the slow pas de deux in the center). Listen to this show sometime. Listen to this amazing music. It has something amazing from every food group and the tunes are freaking haunting. It's so easy to listen to the score and close your eyes and visualize Ray Bolger and Tamara Geve and Helen Broderick (yes, Brodrick Crawford's mom) onstage at The Imperial, before an audience dressed in evening dress and slipping out for a drink at Sardi's during intermission and then off to The Savoy to see Dwight Fiske and his songs at the piano or dance the night away at The Plaza. This show just perfectly captures that amazing era between 1933 and 1941 SO wonderfully.
And the jokes are so modern. A woman sining "I was made, on lemonade, the heart is quicker than the eye." Hilarious.

Since asked.
'Ragtime' with 'Gypsy' sometimes overtaking depending on my mood.
If someone put a gun to my head and made me choose between Sweeney Todd, My Fair Lady, and The Last Five Years, I think I'd go with My Fair Lady. It's just so wonderfully perfect with the contrasting characters and their journeys and the MUSIC! *dies*
Okay, I'm shutting up now.
Hmmmm - I'll have to go with Avenue Q - even though I haven't seen it yet, I JUST LOVE IT!
My favorite one that I have seen is probably The Lion King
Guys and Dolls. I really think it's the perfect musical, as many have said.
Jesus Christ Superstar
A toss-up between Sweeney Todd and Into the Woods.
Cabaret hands down.

Okay, I'll be the one to stand up here. Why not Wicked? We all have our own opinions, and mine is that Wicked and RENT are my favorite shows, is that a problem? I certainly hope not.
Aaaaaaanyway. RENT is an extremely close second, with DRS as a third. Oh, you only asked for 1? Sorry, it's hard to pick!
I agree that people should be allowed to pick Wicked if they want to. Is it a rule that musical fans have to dislike Wicked? Anyway right now I have to say Dirty Rotten Scoundrels. In my opinion it is everything a musical should be.
Too many to choose but Wicked is not in the running
