I think unless Neil Patrick Harris is really bad in Hedwig, it's his to lose. Jefferson Mays is probably a worthy contender.
I think Karimloo will get a nod, along with Andy Karl, and either Stephen Pasquale, Zach Braff or Zachary Levi in the fifth slot. Also, they may have one of the gentlemen from If/Then (Anthony Rapp possibly) placed in leading actor. (I doubt it but you never know.)
The part of Hedwig sounds like a huge shoo-in for a Tony if he/she (He's trans, right?) is done right. And NPH is a wonderful actor, so more than likely, NPH will win

I can't imagine Andy Karl getting nominated over Stephen Pasquale. Also, I really think this award is Jefferson's to lose. He got glowing notices in the fall that have yet to be even close to matched by anyone else this season. Also, even if Ramin and Will are good in Les Mis, don't be surprised if they're snubbed (especially Will) to make room for people doing good work in new shows.
Neil Patrick Harris is a VERY talented actor-I've seen him since 2001 (at a regional theater). He was amazing then and continues to impress me. He always puts 110% into a role and really is a chameleon like actor. I think he has consistently worked hard and delivered brilliant performances. I think so long as he can convince that audience he's HEDWIG, he's got it. He's a very strong actor and I think that people forget his heart is with the theater despite his tremendous success on TV.
Also, Norbert MAY be nominated, but I'd bet he's the sole nomination for the show and not very likely to win.
I'd like to think Zachary Levi would get a nod for First Date, even if he's incredibly unlikely to win.

I feel like people never learn from past years. I loved Norbert in Big Fish and Zachary was the only redeeming thing about First Date, but neither of them will be nominated in a year when their shows closed in the Fall and there are actually GOOD performances happening in the Spring. It'd be one thing if there were a lack of quality performances this Spring, but there are a ton of good ones. Between Jefferson Mays, Bryce Pinkham, Stephen Pasquale, Andy Karol, and the performances I haven't yet seen (Ramin, Will, Adam Jacobs, NPH, Zach Braff), I can't imagine that the nominating committee is going to dip into the Fall choices. Yes Rob McClure was nominated last year for Chaplin, but he also received RAVE reviews across the board. And there weren't other actors in the Spring who were well received who could have taken those spots. Neither Butz not Levi received the kind of attention necessary to score a nomination over these other options in such a strong year.
James Iglehart is the clear front runner and likely winner for Best Supporting Actor right now. He just has to hope Disney doesn't try and place him in the leading category. Disney has done this before with supporting roles. For example, Disney petitioned to have Kara Lindsay placed in leading actress category for Katherine Plumber in Newsies despite knowing Audra McDonald and Jan Maxwell were the clear frontrunners. If she was left in the supporting actress category with her glowing reviews Lindsay would have been a frontrunner and strong challenger against Judy Kaye's performance in Nice Work if You Can Get It.
^^If they really put JIM as Leading Actor, that would be completely foolish. There would be no way that he would beat Neil Patrick Harris or Andy Karl for that. If he gets Featured (most likely he will), BOOM! Lock in.
The Disney producers should learn from the Matilda Producers: Don't lose the easiest Tony ovrt ignorance. (Sorry to mention Matilda, but...)
